Characterizing protein-protein interaction regions is an arduous process. Protein painting involves the use of synthetic organic dyes that coat the surface of native preformed complexes. The surface of the protein complex becomes then protease resistant. As a result, only the peptides buried in the "hot spots" inside the protein complex can be be analyzed by LC-MS/MS. This allows for the determination of the amino acid sequences at the interface of protein complexes.
