The FACSJazz cell sorter installed in a specifically designed biological safety cabinet (BSC) allowing us to sort BSL2 samples. The system has 3 laser excitation (405, 488, and 647 nm) with the ability to measure up to 6 fluorescent parameters, 2 light scatter detectors, time, plus Pulse Processing for width and area measurements. The sensitivity is better than 125 MESF-FITC and 125 MESF-PE with less than 1 correlation error per 108 events. The system has a fixed 100 um nozzle making it very stable and with less shear stress on live cells. Sorting at a fixed 27 psi pressure at 39 kHz with an average threshold of 10,000 events per second, a two-way sort achieved a purity of 98% and yield of >80% of Poisson’s expected yield for both populations. Index sorting is also available allowing the review of the complete flow phenotype of every cell sorted in to a multi-position sort device, such as a 96-well tray.
